Effect of amylin and calcitonin gene-related peptide on insulin-stimulated glucose and calcium transport in the diaphragm.

چکیده

Adipocytes were isolated from epididymal fat pads from the same rats by the method of Rodbell [4] and resuspended in Krebs bicarbonate buffer containing 4% (w/v) bovine serum albumin and 0.1 mM-D-glucose. Aliquots of cell suspension were preincubated for either IS min (myocytes) or 10 min (adipocytes) with or without insulin. Transport was started by the addition of [3H]-2deoxy-11-glucose and stopped either after 10 min by addition o f ice-cold phloretin solution (myocytes) or 5 min by centrifugation through dinonylphthalate (adipocytes). Radioactivity was measured in the cell pellets and corrected for trapped extracellular medium.
